What about Prince Sable (For the Frog The Bell Tolls), Lark (Pilot Wings 64), Ray MK III (Custom Robo Arena), Waluigi or even someone really obscure like Bubbles (Clu Clu Land) or Ayumi Tachibana (Famicom Detective Club?)

I dunno about Lark, though aircraft is technically a new diverse potential I guess, it doesn't click to me as much as the other characters you mentioned.

Yeah, the thing is, after seeing Bomberman in SSF2 v0.9, I have to say personally, aside from the bombs and bomb-related moves, I can actually see Lark using most (if not all) of his moveset (I'm not saying Lark would be a clone of Bomberman, all I'm saying is that somehow I can just picture Lark using most of his tilts and smashes and the jet pack. His u-smash could be a double-wing-slash upwards and his d-smash could be something like him spinning along the ground rapidly with the aforementioned pair of wings themselves via his Bird-man suit from Pilot Wings 64. If anyone has any ideas for his specials aside form the jetpack, I'd like to hear it.) Also, can anyone picture a Rhythm Heaven character for SSF2? Preferably, Rhythm Girl (the red-dress girl from RH.)

Gonna go ahead and speculate all the characters that'll possibly get a redesign or slight touch up:

Ness and Peach need those redesigns. Maybe Megaman too but it's not much of a necessity as the other two I mentioned.

Seeing a few people mention the lack of antagonists (I think the only real one so far is Meta Knight) in the game, and also seeing the way this roster is going (with Issac confirmed, also being a protagonist), I speculate that about 90% of the unlockable characters will be antagonists/anti-heroes in reflection to the protagonists (e.g Bowser [Mario], Bass [Megaman], Eggman/Knuckles [Sonic], e.t.c).

Lloyd needs one too. Even if the colors match his sprites doesn't.

Meta Knight has appeared a lot more times as an ally rather than a villain (tough in Revenge of Meta Knight he appears as a full villain; Adventure/Nightmare in Dream Land is debatable, mostly because Dedede wasn't the real villain)
The only one I find as a Villain is Wario, because he always acts as a greedy man, but it's still debatable.
Honestly, I hope they add Eggman as a character, because I see more moveset potential on him than Shadow (not saying that Shadow would be a clone, he obviously got magic, guns and... well I never playedd as Shadow in most Sonic games, don't judge me)

Wario actually also is a protagonist in his own series as well, so technically, along with your point of Meta Knight being more of an anti-hero, we have no full antagonists at all.
I think that major antagonists like Dr Wily and Eggman will more likely become bosses in Adventure Mode, but I do see your point of Eggman having a potential moveset (reflecting since Bowser actually appeared as a full character in Brawl), though I think if there is gonna be a third Sonic character, which is quite unlikely, it's gonna be Knuckles.
Shadow...just no. He's mainly supported by fanboys, and in most Sonic games I don't really find him that special. General reception for him isn't gonna be that great anyway.
I think Bass has potential as being the playable Megaman villan, but I'm sure people are just gonna mark him as a Megaman clone...and Zero's a bit too under the radar.

Can we have another characters added from the Naruto Shippuden series such as Uchiha Sasuke or Hatake Kakashi?

This could be fun if we had more characters!!!

Maybe Mewtwo is more likely now, since they added Silph Co(Team Rocket headquarters). Could be Mewtwo's home stage?

I think any Ray model from Custom Robo would be a strangely different character, the assist trophy of Ray on Brawl was tony because according to the series Custom Robos are actually quite small.

I speculate that Quote from Cave Story will join the fray! He's a great character with a unique selection of techniques/guns/other weapons and very versatile, indeed! His stage would be First Cave or that rabbit village or something of the like. Who's with me? ALSO! Soul and Maka (as one character) from Soul Eater, they were in a Soul Eater game on Wii. Finn the Human is perfectly eligible - appeared on 3DS in his own game.
