My tier list.

S tier
Isaac. Even if he had terrible stats, the D6 would still make him among he strongest in the game. With his good stats combined with the potential to have an insane amount of chances at powerful items on each floor (especially combined with items like the habit) he is easily the strongest character.

A tier
Cain. Good stats. Having only good pills is an utter boon and can make you powerful fairly early on. Can heavily abuse the cups and balls game, so will virtually never run out of supplies and has a great chance at getting skatole.

Judas. Amazing stats and good starting item. Probably the strongest character at the start of the game. Unlike the characters above him, if he comes across a good active item he will either have to sacrifice his starting bonus or skip the item room (Cain can grab the item, Isaac can reroll). His low starting health can also potentially limit his ability to use devil rooms.

B tier
???. Decent stats, very fragile. If he gets one of a few select items, he can be one of the most powerful characters in the game. Somewhat mediocre otherwise.

Maggie. She's gotten better with the other increased focus on sacrificing health in WotL, but she's still kind of bad. Reaaaly slow. Has Judas's active item problem but she's not as good and neither is her item. Safe pick for beginners. She does have the advantage of always having enough hearts to use the devil room, though.

C tier
Eve. Bad stats and underwhelming items. Even her dream build of half a heart with a bunch of soul hearts is cute at best. Her central problem is that she's supposed to be built around the concept of health sacrifice, but it's barely ever worth it to actually do so on purpose. She's not terrible or anything but she's definitely the worst.

Don't know where to put Samson since I haven't played him enough.

Sacrifice/library rooms are usually not worth it.

A lot of the new items are really strong, including some of the ones you can get in shops. Always go into shops if you have 15+ coins (7+ with steam sale).

Along with the secret room (which must be bordering at least 2 other rooms) there is now a super secret room, which will only border a single room. If you have a bunch of extra bombs you can look for this. Usually it's a better idea to go for the normal secret room first though since it's easier to find.

If you get a spirit heart (the white one) to drop, don't pick it up until you've already killed everything on the floor including the boss and are about to go down to the next floor. When you go to a new level with a half spirit heart, it turns into a read heart container.

BTW Samson sucks major a**. He has one heart, terrible stats, and Blood Lust sucks, when he kills someone he gains some damage bonus for the room, BIG WHOOP. Atleast Eve can have plus 3 damage and have it s'long you have half a heart. Blood Lust however can help you somewhat on bosses that spawns enemies but if you got like Larry Jr. You ain't getting no bonus damage, which sucks. Now maybe if you were OG at this game and were awesome and could beat the game without taking damage I could understand but if that were the case, you'd be better off with Eve if you were that good, Samson is basically just for someone who'd like a real challenge, but overall, he sucks, badly.

I think Samson is average.

Blood lust seems strong in certain rooms and, more importantly, on some bosses. Larry Jr and stuff are super easy. He's good on some of the bosses that are actually important (assuming you didn't get OP items) like Mom, Satan, ???, and especially Mom's Heart/It Lives. Usually the bosses that don't summon anything are on the easy side anyway. And one heart isn't that much of a disadvantage really, early levels are easy anyway and you'll get more. The only issue is the potential at missing out on a few devil rooms.

He's definitely better than Eve lol, I'm just not sure how exactly he compares to ??? and Maggie.

Yes habit is really good with yum heart for sure because you can coast until womb but after that you sometimes start to wish you had a more powerful active item that habit would let you spam (depending on your setup). Habit's amazing on pretty much everyone (isaac with habit = wtf) and I still think maggie is very sub-par, especially considering you won't get that every time. You also will have a LOT of trouble on certain parts if you don't get movespeed ups.

Yeah once the damage aura starts kicking in you cannot just ride it, but still, 2 hearts then get another one back, that's sick.

But yeah, Cain is really useful for getting awesome items and if you get the I AM ERROR room with the butt load of pills, you basically win. Also on another note, I'd just like to point out that Technology and A Lump of Coal go hand and hand and makes me cry and smile.

Yo, I hate Chocolate Milk, well I love chocolate milk irl but I hate it as a weapon. I hate to tap the direction I want to go in for non charging, that sucks.

It can be really strong sometimes

But yeah usually it's just annoying, the damage charge should be a bit higher.

Cat's head sucks.

It can be good if you avoid spending them for a few rooms before a boss. He'll just get bursted down to half health. Also one of the stronger habit items for damage imo.

It's usually a lot worse than infestation though. #1 Gurdy strat = infestation + temperance card in boss room.

Just got fetus + poison bombs + mr. mega.

Probably the easiest run I've ever had.
