Well, because buttons can't have animations because of their limited keyframe space, I suggest on the frame you wish to have the animation (up, over, down) you use a movie clip and place the animation in there.
If you put the movie clip of the animation on the Over frame, then when the cursor hovers over it it will play. Does this help?
